Dans l'hotel du Grand-Prieur de Saint-Gilles, est constitué en 1868 le musée grâce au leg des collections et des oeuvres du peintre arlésien Jacque Réattu (1760-1833) propriétaire des lieux depuis 1822 . Les collections sont enrichies en 1941 par les collections de l'ancien musée des Beaux-Arts, créé en 1924 dans le palais de l'Archevêché. Depuis 1948, date de réouverture au public il a été considérablement agrandi et restauré.
